dc.description.abstractIsolasi bakteri dan uji aktifitas enzim amilase kasar termofilik dari sumber air panas Gurukinayan Kecamatan Payung, Kabupaten Karo telah dilakukan di Laboratorium Mikrobiologi Departemen Biologi, FMIPA USU Medan dari bulan Pebruari sampai dengan Juli 2008 Penelitian ini bertujuan untuk mendapatkan dan mengkarakterisasi bakteri termofil yang berpotensi amilolitik serta mengetahui aktifitas enzim amilase kasar. Hasil isolasi mendapatkan 25 isolat penghasil amilase melalui uji iodine, dipilih tiga isolat yang paling besar zona beningnya yaitu GK 4 (35,20 mm) diikuti GK13 (32,18 mm) dan GK14 (30,15 mm). Isolat GK13 menghasilkan amilase yang bekerja optimum pada suhu 60oC dengan menghasilkan glukosa lebih banyak, sebesar 633,65 ?g/ml dan pada pH 6,0 sebesar 639,59 ?g/ml.en_US
dc.description.abstractA study on isolation and activity assay of crude amylase thermophilic bacteria from hot spring Gurukinayan Kabupaten Karo had been carried out in Laboratory of Microbiology Department of Biology, Faculty of Mathematics and Natural Sciences. University of Sumatera Utara from February to July 2008. This study was aimed to obtain and to characterize the amylolytic thermophilic bacteria and to know the activity of crude amylase. Twenty isolates showed amilolytic activity based on iodine test. Three out of twenty were choosen for examination of their crude amylolityc potential based on diameter of clearing zone GK4 (35,20 mm), GK13 (32,18 mm), and GK14 (30,15 mm). GK 13 showed optimum enzyme activity at 60oC by releasing 633,65 ?g/ml glucosa and pH 6,0 by product 639,59 ?g/mlen_US
